Abstract: Aim: Due to the urgent need for anticancer agents, investigation of endophytes medicinal plants growing in special environments is considered a promising approach search bioactive natural products. Materials and Methods: The ethyl acetate extracts cultures twelve endophytic fungi were isolated from Calotropis procera fam Apocynaceae, Solanum nigrum Solanaceae Forsskaolea tenacissima Urticaceae one richest areas plant biodiversity Saudi Arabia, Najran. All identified subjected preliminary screening assays cytotoxic activity. Results: Four (Cladosporium herbarum (1), Hortaea werneckii (2), Penicillium solitum (3) Eurotium chevalieri (4)) exerted activity against HepG2, T-47D, HCT-116 RAW 264.7 MTT cell viability assay. highest effects observed extract on HepG2 lines with IC50 values 13 μg/ml 42 μg/ml, respectively. showed rather weak line 357, 347, 345 420 1,2,3 4, High 1, 2 3 T-47D 78, 69 94 Conclusion: These represent candidates future metabolites potential medical applications. Phytochemical investigations of Calotropis procera leaves have led to the isolation of two new compounds: quercetagetin-6-methyl ether 3-O-beta-D-4C1-galacturonopyranoside (3) and (E)-3-(4-methoxyphenyl-2-O-beta-D-4C1 -glucopyranoside)-methyl propenoate (4), along with eleven known metabolites: nine flavonol and two cinnamic acid derivatives. All metabolites were isolated for the first time fro...
